TransIT® In Vivo Gene Delivery System
Features and Advantages
- Efficient and safe non-viral delivery of exogenous DNA complexes into tissues of live mice
- Easy-to-use with two components only
- Enables in vivo protein production
- Allows organ-specific gene expression studies
- Supersedes potentially harmful virus systems
- Injection into the mouse tail vein
- Broad tissue-expression range
Product Description
The TransIT® In Vivo Gene Delivery System is the first product specifically designed to introduce nucleic acids into live mice, allowing for organ-specific genetic studies that were previously impossible to perform on a routine basis. This product contains a unique polymer formulation that facilitates efficient delivery of exogenous DNA complexes into tissues of live mice without the use of potentially harmful viruses. The easy-to-use system consists of two components: a proprietary polymer that forms complexes with the DNA of interest, and a dilution/injection reagent for delivering the DNA into the animal. The DNA is injected into the tail vein of the mouse and target organs are assayed using simple reporter gene assays. The product yields a broad tissue-expression range. |

TransIT® -TKO: small interfering RNA (siRNA) Transfection Reagent
|
|
|
|
| |
For high-efficient small interfering RNA transfection
Features:
- Delivers siRNA targeted against both transient transfected and stable genes in a variety of cell lines while not effecting untargeted expression of similar genes - significantly reduced cell damage
- Ideal for all siRNA-mediated gene silencing studies
- No media change or serum addition required
Product Description
The novel TransIT® -TKO reagent enables highly efficient siRNA transfection with significantly reduced levels of cell damage as compared to cationic-liposome based transfection reagents. Transfections are most effective when carried out in complete growth media without a media change or serum addition. When complexed with even low levels of siRNA, TransIT® -TKO reagent successfully knocks out target gene expression in a variety of cell lines. Transgene expression can be silenced either when the siRNA is transfected before, concurrently or after the transfection of the expression vector. These unique features make TransIT® -TKO transfection reagent ideal for all siRNA-mediated gene-silencing studies. It is the only transfection reagent designed to efficiently deliver small interfering RNA to cells in vitro. TransIT® -TKO reagent, combined with the correct siRNA sequence against the target mRNA, can silence endogenous, stable or transient gene expression.

Figure 1. Achieve high antibody titers using the TransIT-PRO™ Transfection Kit in suspension CHO cells. Human IgG1 was produced by transient transfection using TransIT-PRO™ and PRO Boost Reagent (1:1:1), 25 kDa linear PEI (6:1) or FreeStyle™ MAX (1:1) transfection reagents according to the manufacturers or published protocol (reagent:DNA ratio). Transfections were performed using 1 µg plasmid DNA per milliliter of culture and 0.5 x 106 cells/ml at the time of transfection. FreeStyle™ CHO-S cells were cultured in 20 ml of FreeStyle™ CHO Expression medium in 125 ml shake flasks. (A) Day 3, 5 and 7 supernatants were clarified and analyzed using a human IgG-Fc sandwich ELISA. Error bars represent the standard deviation of triplicate technical replicates, 25kDa linear PEI is duplicate technical replicates. (B) Day 7 supernatants were clarified and analyzed by Western blot. An IgG standard was included for quantification estimate (S1= 1.6 mg/l, S2= 3.2 mg/l, S3 = 6.3 mg/l).

(A) Luciferase expression was assessed 48 hours post-transfection using a conventional luciferase assay. Error bars represent the standard deviation of duplicate wells. (B) Human IgG1 was quantitated from day 5 clarified supernatants and analyzed by a human anti-Fc sandwich ELISA. Error bars represent the standard deviation of triplicate wells.

TransIT-TKO® HTS-96 Plates
|
|
| |
Features and Advantages
- Ideal for screening applications: High efficiency knockdown in a 96-well plate format allows rapid, simultaneous screening of multiple siRNA conditions including siRNA concentration, sequence and cell type in one plate.
- Customized: Plates are available in four working concentrations of TransIT-TKO® Reagent for optimization with a wide range of cell types. The Titration Plate is available for testing all four levels at one time in a preliminary screen.
- Convenient: Easy to use, serum compatible protocol; can be performed in a single 96-well plate or using a two step transfer method.
- Consistent performance: Efficient and reproducible siRNA delivery among multiple wells in a plate for performing replicate samples.
Product Description
The TransIT-TKO® Reagent is supplied as a dry film on clear plastic, sterile, 96-well plates which have flat-bottom wells suitable for cell culture. Each plate is supplied individually vacuum sealed in a foil pouch to maintain sterility and ensure effectiveness. Researchers can test up to eight different siRNA sequences in triplicate format, at four different levels of reagent, from L1 (lowest) to L4 (highest), using the TransIT-TKO® HTS-96 Titration Plate (MIR 2530), on their particular cell type. Once the optimal level has been determined based on the highest efficiency of transfection with the lowest cellular toxicity, additional plates can be purchased at the desired level. Transfections can be performed using a single TransIT-TKO® HTS-96 Plate or using a two step transfer method, where the TransIT-TKO® HTS-96 Plate is used as a reagent plate to form complexes and transfer to the plate of choice.

The TransIT®-mRNA Transfection Kit is optimized for efficient delivery of various types of RNA to mammalian cells. Both mRNA and viral RNA of various lengths can be delivered effectively to a wide range of cell types. In addition, transfection of mRNA leads to high level protein expression with minimal cytotoxicity. As an added benefit, this kit has been optimized for use in the presence of serum containing media thus eliminating media changes or serum addition. MIR 2250 provides sufficient reagents to perform up to 500 transfections per well of a 12-well plate.
Cell lines successfully transfected: A549, CHO-K1, COS-7, HEK 293, HeLa, HepG2, NIH 3T3, and Vero cell lines.

Product Description
TransIT®-siQUEST™ Transfection Reagent enables highly efficient siRNA transfection with significantly reduced levels of cell damage as compared to cationic-liposome based transfection reagents. Transfections are most effective when carried out in complete growth media, with no media change or serum addition required. TransIT®-siQUEST™ Reagent efficiently delivers siRNA to a wide range of cell lines, promoting high levels of target gene expression knockdown. These unique features make the TransIT®-siQUEST™ Transfection Reagent ideal for siRNA-mediated gene silencing studies. The TransIT®-siQUEST™ Reagent is a complement to Mirus' popular TransIT-TKO® siRNA Transfection Reagent. Each unique formulation provides a distinct transfection profile for high efficiency, broad-spectrum siRNA delivery.
Cell Lines Successfully Transfected: A549, CHO-K1, COS-7, HEK 293, HeLa, Hepa1c1c7, HepG2, MCF-7, NIH 3T3, RAW 264.7, and Vero cells.
